OriBioTM Pfu DNA Polymerase is a Pyrococcus furiosis clone of DNA polymerase gene expressed in E.coli . The enzyme catalyzes the template-dependent polymerization of nucleotides into duplex DNA in the 5´→3´ direction. Pfu DNA Polymerase also exhibits 3´→5´ exonuclease (proofreading) activitythat enables the polymerase to correct nucleotide incorporation errors. It has no 5´→3´ exonucleaseactivity. Pfu DNA polymerase is 6 times more accurate than Taq DNA polymerase. The enzymeis highly thermostable (90% active after 1 hour @ 95℃), and has the capability of incorporatingmodified nucleotides. Pfu DNA polymerase generates blunt-end PCR products, which cannot be used directly for a TA cloning reaction.
