OriBioTM 2× HiFiTaq PCR Mix with loading dye is supplied as a 2× concentration mixture containing a blend of HiFiTaq DNA Polymerase, dNTPs, Mg2+, reaction buffer and stabilizer. It is generally used at 1× concentration with DNA template and primers in a total reaction volume of 20 or 50 μl. The fidelity of HiFiTaq DNA polymerase (a mixture of recombinant Taq polymerase and a high fidelity proofreading DNA polymerase of choice) is approximately 4 to 6 times that of Taq DNA polymerase. It is suitable for amplification of DNA fragments up to 10 kb in length with relatively higher fidelity than using Taq DNA polymerase alone. Its benefits include quick and easy setup, high sensitivity, specificity, and good stability, minimizing manual error, time saving, contamination reduction, and is ideal for high-throughput gene detection and rapid clone screening. The PCR products generated using 2× HiFiTaq PCR Mix contain dA overhangs at the 3´end, and are suitable for downstream TA cloning. The dye-containing end products after PCR reaction can be loaded directly for gel electrophoresis and detection, and can be purified for subsequent experiments. The red dye contained migrates at a rate that is close to a double-stranded DNA fragment of 300 bp in a 1% TAE agarose gel.
